Programme

Role of chromatography in analytical chemistry. Classification of chromatographic systems based on the quality stationary and mobile phase.

Basic definitions used in chromatography and their relations in the separation.

Role of retention, kinetic efficiency and selectivity in the separation.

Prerequisites for gas chromatographic analysis. Relation of vapor pressure and retention in gas chromatography. Efficiency differences of packed and capillary columns and their applications. Basics of gas-solid and gas-liquid chromatography and their applications.

Mobile phases in gas chromatography. Effect of mobile phase quality on the separation. Van Deemter and Golay equations and thier explanations.

Stationary phases and their characterisation. Choosing the appropriate stationary phase based on analyte structure and boiling point.

Instrument design. Packed column and capillary column gas chromatographs. Instrument parts and their function.

GC detectors: operation, classification, analytical characterisation.

Qualitative and quantitative analysis in gas chromatography.

Basic theory of liquid chromatography. Parameters influencing resolution. Application of Van Deemter and Knox equations in LC. Classification of fluid chromatographic techniques based on the forces resulting solute migration. Basics of kinetic theory. Recent developments in liquid chromatography.

Characterization of packed columns. Effect of particle diameter and its distribution on resolution. Typical mobile phases and their effect on resolution.

Normal phase liquid chromatography (NP-HPLC). Packings: silicagel, aluminum-oxide, modified silicagels. Mobile phases. Parameters influencing retention, selectivity and resolution.

Reversed phase liquid chromatography (RP-HPLC). Packings: silicagel based, carbon based and other types. Effect of mobile phase composition, pH, ionic strenght and temparture on the separation.

Application of secondary equilibria for increased selectivity. Ion-supression and ion-pair chromatography in RP-HPLC.

Basics of ion-exchange chromatography (IE-HPLC). Organic and inorganic packings an their applications.

Methodologies for the separation large molecules. Size exclusion chromatography: gel chromatography, gel permeation.

Basics of gradient elution.

Instrumentation in HPLC: pumps, detectors, system control

Important applications and some relating sample preparation methods (LLE, LSE, SFE)

Recent developments in liquid chromatography. Capillary columns and monolith packings. Hyphenated techniques (e.g. LC-MS)

The subject lays emphasis on the basics and applications of chromatographic analysis: theoretical background and practice will be discussed in order to develop skills for method development and application of hyphenated techniques.

Requirements: Fullfillment of the following tasks: literature search, design and cost estimation of the analysis, sampling, sample preparation, measurement, evaluation and reporting. Presenting a talk on the project and writing a test. Re-takes: The test can be repeated. Consultations: By previously fixed appiontment with the lecturer.
